Tools and Innovative Technologies for Drug DiscoverySci-Tec, Inc. is developing a new class of pharmaceutical agents for modulating enzyme activity: "on-Site Assembled Pharmaceuticals (SAP)". The SAP is a combination of two or more pharmaceutical compounds which can bind to the same active site of the target enzyme. When bound to the target, the compounds form a stable molecular complex at the active site. The distinctive element of the approach is that the active site of the target enzyme plays a critical role in assembly of the molecular complex. The formation of SAP complex only in the presence of the target enzyme is beneficial for improving specificity and reducing toxicity of drugs. The additional advantage of the SAP approach is that a large molecular complex, which "violates" Lipinski’s Rule-of-Five now can be assembled inside cells from much smaller Ro5-compliant molecular fragments. |
